摘要
目的体外分离培养和鉴定小鼠脂肪源性干细胞,向成骨方向分化,为后续的实验选取合适的种子细胞奠定基础。方法从白色脂肪组织中分离培养脂肪源性干细胞,应用流式细胞仪分析其表面的标记。分别通过碱性磷酸酶、茜素红和油红-O染色方法染色方法鉴定向成骨和成脂方向分化。结果脂肪源性干细胞高表达表面标记CD29、CD44,并且能够向成骨和成脂方向分化。结论利用胶原酶消化法,在小鼠脂肪组织中成功分离、培养出脂肪源性干细胞,获得的细胞具有多向分化潜能,为后期组织工程技术修复骨组织,提供了坚实的实验基础。
Objective To isolate, culture and identify adipose-derived stem cells(ADSCs) from mouse and differen- tiate into bone so as to provide a theoretical basis for later experiments which are aim to select suitable seed cells for bone repair. Methods Adipose-derived stem cells were isolated and cultured from white adipose tissue, and surface antigens including CD44 were selected. CD29 and Sca-1 were analyzed by flow cytometry. Differentiation towards adipocyte and osteocyte were determined with different staining methods of alkaline phosphatase, Alizarin red stain and oil-O. Re- sults ADSCs were positive for the pluripotent markers of CD29, CD44 and Sca-1, and able to finish osteogenic and adipogenic differentiation. Conclusions ADSCs can been succeeded to isolate and cultured through the method ofcol- lagenase from mouse adipose tissue. The obtained ADSCs could differentiate into diverse cell types. It provides a theoretical basis for later experiments which are aim to repair bone damage tissue by using tissue engineering.
