摘要
目的 观察全脑缺血再灌注时脑组织抗凋亡基因 bcl- 2蛋白表达及降钙素基因相关肽 (CGRP)对 bcl- 2蛋白表达的影响。方法 应用颈动脉负压分流法制备大鼠全脑缺血再灌注模型,应用免疫组织化学法检测全脑缺血 30min再灌注 3h、 6h以及经颈动脉注射 (CGRP)后再灌注 3h、 6h大脑皮层和海马区 bcl- 2蛋白的表达。结果 大鼠全脑缺血 30min再灌注 3h、 6h大脑皮质及海马 bcl- 2蛋白表达明显增加,应用 CGRP再灌注 3h、 6h大脑皮质及海马区 bcl- 2蛋白表达进一步增加。结论 bcl- 2基因在缺血性脑损伤中起保护作用, CGRP的神经保护作用可能与其上调 bcl- 2蛋白表达有关。
Objective To observe the expression of bcl- 2 protein and the effect of calcitonin gene related peptide(CGRP)on it in brain tissue during whole cerebral ischemia/reperfusion in rats.Methods The model of whole cerebral ischemia/reperfusion was established by method of carotid negative pressure shunt in rats.Immunohistochemistry method was used to detect the expression of bcl- 2 in cortex and hippocampal in control group,whole cerebral ischemia 30 minutes/reperfusion 3 hours group(I/R 3h)、 6 hours group(I/R6h),I/R3h plus injecting CGRP into carotid group(I/R3h+ C)and I/R6h plus injecting CGRP into carotid group(I/R6h+ C). Results bcl- 2 protein expression increased significantly in cortex and hippocampal in I/R3h and I/R6h and further in I/R3h+ C and I/R6h+ C. Conclusion Bcl- 2 gene might play a protective role in ischemia/reperfusion brain injury.The protective effect of CGRP might be related to its upregulating effect on bcl- 2 protein.
