摘要
本文对我国特产资源生漆液中漆树酶的固定化进行了研究。考查了各种无机盐溶液处理不同无机载体,活化反应及漆树酶固定化条件对固定化漆树酶性能的影响。研究表明,经ZrCl_4处理后的多孔硅胶作载体的固定化漆树酶活力回收高、稳定性好、重复使用20次其保留活力仍有75%。同时对固定化漆树酶和自由态漆树酶的反应pH、温度、K_m值作了比较。对漆树酶通过过渡金属氢氧化物固定在无机载体土的机理也作了初步探讨。
Immobilization of Rhus laccase (EC 1.10.3.2) from sap of lac tree was studied. Conditions of various transition metal salts activating inorganic carriers and immobilization of Rhus laccase influence the properties of immobilized Rhus laccase. A porous silica treated by ZrCl_4 solution used as carrier of Rhus laccase immobilization was produced, the activity recovery and stability of the immobilized Rhus laccase is superior to that reported in literature, the retained activity was about 75% after it had been used for 20 times. The chemical and physical properties of immobilized Rhus laccase such as optimum pH, temperature, Km were determind and compared with those of native Rhus laccase. Machanism of the process was deduced from the experimental results.
