摘要
目的 阐明急性压力超负荷后心肌细胞内cAMP浓度升高和心肌碱性成纤维细胞生长因子 (bFGF)变化之间是否存在内在因果关系。方法 用腹主动脉缩窄的方法建立急性压力超负荷大鼠模型 ,检测 6个时相点 (0 5h ,4h,1d ,5d ,10d ,30d)的平均动脉压 (MAP)、左室收缩压 (LVSP)、左室重量指数 (LVMI) ;同时分别用免疫组织化学、原位杂交法检测bFGF蛋白及mRNA表达变化 ,放射免疫法检测心肌cAMP含量 ;并在心肌细胞培养的基础上 ,用异丙肾上腺素 (ISO)刺激培养心肌细胞 ,升高细胞内cAMP浓度后检测bFGF蛋白及mRNA表达变化。结果 发现心肌组织中cAMP、bFGFmRNA及蛋白表达分别于术后 0 5h、1d、5d开始显著增加 ,5~ 10d达峰值 ,术后 30d时均降至正常 ;0 0 1μmol/LISO即可刺激心肌细胞使bFGFmRNA及蛋白表达显著增加。
Objective To investigate the relationship between cAMP and myocardial basic fibroblast growth factor(bFGF) after acute pressure overload.Methods Wistar rats underwent partial occlusion of the abdominal aorta(OG) or a sham procedure(CG).At 0.5h,4h,1d,5d 10d,30d after operation,left ventricular mass index(LVMI)、mean arterial pressure(MAP) and left ventricular systolic pressure(LVSP) were detected.In addition,the expression levels of bFGF mRNA、bFGF protein and cAMP concentration were studied by in situ hybridization、immunohistochemistry and Radioimmunoassay (RIA).The cardiomyocytes were cultured to observe whether bFGF gene expression can be induced by cAMP.Results It was found that cAMP concentration、bFGF mRNA and protein expression were increased significantly at 0.5h、1day、5day after acute pressure overload,reached the peak at 5 day 10 day and returned to normal at 30day after operation;Elevation of cAMP concentration in clutured cardiomyocytes stimulated by isoproternol(ISO,0.01umol/L) increased bFGF mRNA and protein expression in cultural medium.Conclusion These results suggest that cAMP plays the role in the mechanism of bFGF gene expression induced by pressure overload.
