姬松茸多糖对小鼠骨髓来源树突状细胞抗肿瘤作用的影响 被引量:5

摘要 目的:研究姬松茸多糖对小鼠骨髓来源树突状细胞抗肿瘤功能的影响,为临床应用提供理论依据。方法:体外培养小鼠骨髓来源DC,加入ABP溶液,流式细胞仪检测DC表面CD86和CD11a的表达,混合淋巴细胞反应检测ABP对DC抗原提呈能力的影响,MTT法检测ABP对细胞毒性T淋巴细胞(CTL)对H22肝癌细胞杀伤功能的影响。结果:中剂量ABP组(100μg/mL)DC表面CD86、CD11a表达上调;CTL对肿瘤细胞的杀伤率升高,差异有统计学意义(P<0.05)。结论:适当浓度的ABP能促进DC分化成熟,增强DC的抗肿瘤作用。 Objective: To observe the regulation on the functional of dendritic cells (DC) derived from murine bone-marrow by Agaricus blazei polysaccharides (ABP) in vitro and provide the experiment basis for clinical application. Methods: Different dosages of ABP were added to the culture medium, the expression of CD86 and CDlla were analyzed with a flow cytometer; Killing capa- bilities of allogeneic T cells induced by DC were examined with MTT method. Killing capability of cytotoxicity T lymphocyte on H22 hepatocellular carcinoma ceils was examined with MTT. Results: The expression of CD86 and CDlla as well as killing capability of eytotoxicity T lymphocyte increased in the middle dose of ABP group, with significant difference (P〈0.05). Conclusions: The suitable concentration of ABP can promote the maturation of DC and enhance the antitumor effect of DC.
