Cryopreservation and homografting of fetal brain tissue in rats

摘要 目的 探讨冻存大鼠胎脑细胞体外培养的活性及其移植后的存活情况。方法 对经冷冻保存 1~ 2 4周的WK大鼠胎脑细胞进行体外培养 ,观察其存活情况 ,并将保存 2 4h后培养 14d的胎脑细胞行脑内定位移植。结果 冻存 1~ 2 4周的胎脑组织其活细胞率在 90 %以上 ,复苏后培养 14d的胎脑细胞活性最强 ;胎脑细胞移植后可在受者体内存活至少 30d以上。结论 冷冻保存后培养的胎脑细胞能够存活 。 Objectives To study the living character of cryopreservation of in vitro cultured fetal brain tissue in rats and the survival of experimental allograft.Methods The WK rat fetal brain tissue for cryopreservation of 1 to 24 weeks was cultured in vitro to observe the survival of the allograft. The rat fetal tissue with 24 h cryopreservation and 14 day culture was transplanted to homogeneous rat caudatum. Results The cryopreserved rat fetal brain tissue cultured in vitro could had a living cell rate of over 90?%. The livingness of the fetal brain cells cultured for 14 days was the strongest. The rat fetal brain tissue could live more than 30?days after allografting and could not be rejected. Conclusions The cryopreserved technique for fetal brain tissue provides an efficient and reliable storage method for programmed brain transplantation operation and other basic medical research.
