4种根管封闭剂体外细胞毒性的比较 被引量:4

摘要 目的:评价4种根管封闭剂的生物相容性。方法:本实验通过X射线能谱仪对MTA、AH Plus、Roeko Seal、Cortisomol 4种根管封闭剂进行元素分析,并采用CCK-8法检测各材料凝固后即刻和1周时在100%和50%两种浸提液浓度下对L-929细胞的体外细胞毒性作用。结果:同一固化时间组内两种浸提液浓度相比,AH Plus组无论是固化即刻组还是固化后1周组,当其浸提液浓度为100%时,1、2、4 d各时间点的细胞增殖率均低于50%浸提液(P<0.05),而其他3种封闭剂的即刻组和1周组样本中100%与50%浓度浸提液相比,各时间点的细胞增殖率均无显著性差异(P>0.05)。同一时间点内各封闭剂相比,无论是固化即刻还是固化后1周组,其浸提液浓度无论是100%还是50%,各时间点的细胞增殖率由高至低依次为MTA组、Roeko Seal组、AHplus组、Cortisomol组;MTA组与Roeko Seal组相比P>0.05,其他各组间比较P<0.05。结论:MTA与Roeko Seal的细胞毒性最小,AH Plus的细胞毒性居中,Cortisomol的细胞毒性最大,但毒性级均未超过3级。 AIM: To detect the cytotoxic effects of MTA, AH Plus, Roeko Seal and Cortisomol on L-929 cell line. METHODS : Element analysis of MTA, AH Plus, Roeko Seal and Cortisomol were conducted by energy dis- persive X-ray spectrometer. CCK-8 was used to detect the in vitro cytotoxic effects of the extractions at 100% and 50% after immediate solidification or 1 week after solidification of the sealers on L-929 cells. Cytotoxicity was evalua- ted by relative growth ritio (RGR) standard. RESULTS: The 4 root canal sealers were composed with different Ele- ments. Cell growth in 100% AH Plus extract group was lower than that in 50% group at all time points ( P 〈 0.05), while no significant difference was observed between the 100% and 50% extracts of the other three sealers (P 〉 0.05 ). At the same time points, MTA group showed the highest cell proliferation, followed by Roeko seal, AHplus and cortiso- mol group. No significant difference was found between MTA and Roeko Seal groups(P 〉 0.05 ). However, significant cell growth difference was observed between each 2 of the other groups (P 〈 0.05 ). CONCLUSION : MTA and Roeko Seal have the lowest cytotoxicity, AH Plus has moderate, and Cortisomol has the highest cytotoxicity to L-929 cells. However, the cytotoxicity of all four sealers was less than grade 3.
