小型猪牙髓、牙周膜干细胞的生物学性能比较 被引量:3

摘要 目的:比较小型猪牙髓干细胞(DPSCs)和牙周膜干细胞(PDLSCs)的生物学性能。方法:有限稀释法克隆培养同一个体小型猪的DPSCs和PDLSCs,流式细胞仪检测细胞表面标记物,通过克隆形成率和MTT比色法检测2种细胞的增殖活性;成脂、成骨诱导培养检测细胞的分化能力,Real-Time PCR检测比较2种细胞的成骨相关基因Runx2、Col-1、OCN mRNA的表达水平。结果:小型猪DPSCs和PDLSCs均阳性表达CD146、Stro-1、CD29、CD90,阴性表达CD45;DPSCs和PDLSCs的克隆形成率分别为15.3%和14.6%(P>0.05);2种细胞均具有较强的增殖活性,都具有成脂、成骨分化的能力,成脂能力2种细胞相比无明显差异(P>0.05),体外形成钙化结节的能力PDLSCs强于DPSCs(P<0.05)。PDLSCs中的各成骨相关基因Runx2、Col-1、OCN mRNA的表达水平高于DPSCs(P<0.05)。结论:小型猪恒牙DPSCs和PDLSCs均具有较强的增殖和多向分化的潜能;但DPSCs的骨向分化潜能低于PDLSCs。 AIM: To compare the biological characteristics of dental pulp stem cells (DPSCs) and perio- dontal ligament stem cells (PDLSCs) of miniature pig. METHODS: DPSCs and PDLSCs were isolated and cultured from the same donor by limited dilution method and then their surface antigens were examined by flow eytometry. The cell proliferation was studied by clone formation assay and MTT assay. The cell differentiation was evaluated by osteo- genesis-inducing and adipo- inducing assay respectively. The expression of osteogenesis related genes, such as runt-related transcription factor-2 (Runx2), collagen type 1 (Col-1), and osteocalein (OCN) was detected by Real- Time PCR. RESULTS : The DPSCs and PDLSCs of miniature pigs were positive for CD146, Stro-1, CD29 and CD90, while negative for CD45. The clone formation rates of DPSCs and PDLSCs were 15.3% and 14. 6% respectively (P 〉 0.05). The two kinds of cells showed active proliferation. Both cells could be induced to differentiate into osteo- blasts and adipoblasts. Stronger expression of Runx2, Col- 1 and OCN was found in PDLSCs than in DPSCs ( P 〈 0.05 ). CONCLUSION : The DPSCs and PDLSCs of miniature pig have similar proliferative activity and adipo- genie differentiation potential, however, PDLSCs have greater osteogenic differentiation potential than DPSCs.
