槟榔碱诱导上皮细胞凋亡相关蛋白caspase-3,8,9、Bcl-2和Bax的表达 被引量:8

摘要 目的:观察槟榔碱刺激上皮细胞后凋亡相关蛋白caspase-3,-8,-9、Bcl-2和Bax的表达情况。方法:0,25,50,75,100,125μg/mL槟榔碱刺激上皮细胞Hacat 24 h,Western Blot法检测caspase-3,8,9、Bcl-2和Bax的表达情况。结果:随着槟榔碱浓度的增加,caspase-3、caspase-8表达量逐渐增强,但casapse-9表达未见明显上调;胞内Bcl-2表达受到明显抑制,Bax在槟榔碱浓度为125 g/mL时,可见表达明显上调。结论:槟榔碱可通过内源性和外源性途径诱导上皮细胞Hacat发生凋亡,但高浓度槟榔碱对外源性凋亡途径的活化更为显著;槟榔碱可上调促凋亡蛋白Bax的表达并下调抗凋亡蛋白Bcl-2。 Objective:To study levels of expression of the proteins caspase-3,-8,-9,Bcl-2 and Bax in Hacat cells induced by arecoline.Method:Hacat cells were incubated with different concentrations of arecoline (0,25,50,75,100 or 125 μg/mL) for 24 h.This was done to detect the levels of expression of caspase-3,-8,-9,Bcl-2 and Bax in Hacat cells using Western blot.Result:With the increase in the concentration of arecoline,the expression of caspase-3 and-8 gradually increased,but casapse-9 failed to be significantly upregulated,Hacat cells expressing Bcl-2 was inhibited,whereas Bax was significantly up-regulated when the concentration was 125 g/mL.Conclusion:Arecoline is able to induce Hacat cell apoptosis by endogenous and exogenous pathways,but the exogenous pathway of apoptosis is more significantly activated when the concentrations of arecoline is high.The arecoline can raise pro-apoptotic protein Bax and down the antiapoptotic protein Bcl-2.
