白细胞介素-17对系膜细胞功能的影响及其分子机制 被引量:3

Influence of IL-17 on mesangial cell and its molecular mechanism

摘要 目的研究白细胞介素(IL)-17对系膜细胞增殖及分泌功能的影响,并探讨其可能的分子机制。方法四甲基偶氮唑盐比色(MTT)法检测IL-17对系膜细胞增殖的影响;酶联免疫法(ELISA)检测IL-17对系膜细胞分泌IL-1β的影响;免疫细胞化学法检测系膜细胞NF-κB的表达。结果不同浓度IL-17在不同时间点对系膜细胞增殖的影响均无统计学意义(P>0.05);IL-17可促进系膜细胞分泌IL-1β,且呈浓度时间依赖性,差异有统计学意义(P<0.05);同时IL-17诱导组NF-κB的表达高于空白对照组,差异有统计学意义(P<0.05)。结论 IL-17可能部分通过NF-κB途径引起系膜细胞分泌炎症因子IL-1β,加重肾脏局部的炎症状态,导致肾脏损伤。 Objectives To investigate the effect of IL-17 on proliferation and secretion of IL-1βin the human mesangial cell line (HMCL) and explore the molecular mechanism. Methods The proliferation of HMCL induced by IL-17 was detected by MTT, the secretion of IL-1βin HMCL was measured by ELISA and the expression of NF-κB was detected by immunocyto-chemistry. Results IL-17 did not impact HMCL proliferation (P&gt;0.05) but dose-and time-dependently promoted the secretion of IL-1β(P&lt;0.05). And the expression of NF-κB induced by IL-17 increased comparing to that in the control group (P&lt;0.05). Con-clusions IL-17 may induce HMCL to secrete IL-1βvia NF-κB pathway that results in kidney injury.
