依达拉奉对脑缺血再灌注大鼠的脑保护作用及对过氧化物酶体增殖受体γ(PPARγ)表达的影响

摘要 目的研究依达拉奉对大鼠脑缺血再灌注脑组织的过氧化物酶体增殖受体γ(PPARγ)表达的影响,探讨依达拉奉对缺血再灌注后脑的保护作用。方法 24只Wistar大鼠随机分为对照组、模型组及高、低剂量组,对照组只暴露一侧颈总动脉后缝合,不予任何处理;模型组及依达拉奉高、低剂量组应用改良线栓法制备成功后立刻分别应用相应药物腹腔注射,在24h、72h、7d时间点分别取脑组织,通过免疫组化的方法检测PPARγ的表达变化。结果依达拉奉高、低剂量组PPARγ蛋白表达较模型组明显增多(P<0.05),依达拉奉高、低剂量组PPARγ蛋白表达比较有显著性差异(P<0.05)。结论依达拉奉可通过上调PPARγ的表达来降低炎症反应,且随剂量的加大,上调幅度增加,抑制脑缺血再灌注周围神经细胞凋亡,促进细胞代谢,从而对脑缺血再灌注大鼠大脑起保护,为脑梗死的神经保护治疗从理论上提供支持。 Objective To research the expression of Yidalafeng on PPARγ and its cerebral protection in the rat with cerebral ischemia reperfusion. Methods The animals were randomly divided into contrast group, model group,and high, low dosage of Yidalafeng groups, the contrast group was done nothing, the others of middle cerebral artery ischemia reperfusion were established by suture method. Model group, high and low dosage of Yidalafeng groups were treated with corresponding drugs by intraperitoneal injection. The brain tissue was gotten at 24h,72h,7days to detect the expression of PPAR), by immunohistochemistry. Results Compared the operated groups to the contrast group, PPARγ protein expression was significantly increased in the surgery group. The expression of PPARγ protein was obviously increased in high and low dosage of Yidalafeng groups compared with the model group(P〈0.05). The difference in the expression of PPARγ between high and low dose of Yidalafeng groups was significant(P〈0.05). Conclusion Yidalafeng has pretty good protection on cerebral protection in the rat with cerebral ischemia reperfusion through increasing the expression of PPARγ, and the more dose, more effective.
