福尔马林足底炎性疼痛大鼠模型的建立及其背根神经节辣椒素受体表达水平 被引量:5

Establishment of rat model with inflammatory pain induced by formalin and the expression level of capsaicin receptor in dorsal root ganglion

摘要 目的 评价福尔马林足底炎性疼痛大鼠模型的疼痛行为学表现,并对该模型大鼠背根神经节辣椒素受体表达水平进行分析.方法 SD大鼠24只,随机分为2组,即对照组(n=12)及福尔马林模型组(n=12).对照组不予处理,福尔马林模型组于左后足底注射5%福尔马林0.1 mL,观察造模后大鼠疼痛行为学反应,并以Dubuisson评分方法进行记录比较;2组大鼠在指定时间点取出L3-6节段背根神经节,运用RT-PCR、蛋白质印迹(Western blot)检测辣椒素受体在大鼠背根神经节中的表达水平.结果 福尔马林模型组大鼠呈典型的双相伤害性行为反应,大鼠疼痛反应120 min内积分在5~1,对照组无疼痛反应.福尔马林模型组大鼠背根神经节辣椒素受体表达水平显著增加,与对照组比较有显著性差异(P<0.05).结论 福尔马林足底炎性疼痛大鼠模型可以产生持续的、可信的痛觉过敏,并诱导大鼠背根神经节辣椒素受体表达水平增加,是研究组织损伤诱发炎性疼痛机制的较佳模型选择. Objective To evaluate the behavioral manifestations of inflammatory pain in rat model injected with formalin in plantar,and analyze the expression level of capsaicin receptor of dorsal root ganglion in rat.Methods 24 SD rats were randomly divided into model group and control group.12 SD rats in model group were injected with 0.1 mL 5% formalin in the left plantar,other 12 SD rats in controls did not be treated.The pain behaviors were observed,and Dubuisson score method was used.The segments of L3-6 dorsal root ganglion have been taken from rats in a specific time.The expression level of capsaicin receptor of dorsal root ganglion was detected by RT-PCR and western blotting.Results Formalin injected group showed a typically biphasic nociceptive responses,and had scores from 5~1 in pain response within 5 min(P<0.05).Control group did not have pain response.The expression level of capsaicin receptor of model group was significantly higher than that of the control group (P<0.05).Conclusion The rat model with inflammatory pain induced by the plantar injection of formalin has sustained,credible hyperalgesia,and has an increased expression level of capsaicin receptor of dorsal root ganglion.It might be a good model for the mechanism study of the inflammatory pain induced by tissue damage.
