穿心莲内酯对脓毒症小鼠淋巴细胞免疫功能的调节作用 被引量:5

摘要 目的探讨穿心莲内酯对脓毒症小鼠淋巴细胞免疫功能的影响。方法采用盲肠结扎穿孔(eecalligationandpuncture,CLP)制作脓毒症模型。48只C57BL/6小鼠按完全随机法分为4组,每组12只:假手术组(Sham组)、脓毒症对照组(CLP组)、二甲基亚砜(dimethylsulfoxide,DMSO)组和穿心莲内酯组(AND组)。各组小鼠术后1、6h和12h腹腔内分别注射等体积的生理盐水、DMSO或穿心莲内酯。术后24h流式细胞仪检测脾脏CD4+T淋巴细胞、CD8+T淋巴细胞、CDl9+B淋巴细胞以及CDlle’DC细胞数量;流式细胞仪检测胸腺CD3+T淋巴细胞凋亡情况。结果穿心莲内酯治疗后,小鼠脾脏CD4+T淋巴细胞、CD8+T淋巴细胞、CDl9+B淋巴细胞以及CDllc’DC计数分别为(8.03±0.62)×105/ml、(3.42±0.58)×105/ml、(6.48±1.13)×106/ml、(5.50±0.79)×106/ml,显著高于CLP组(P〈0.01);治疗后的小鼠胸腺CD3+T淋巴细胞的凋亡明显减少(P〈0.01)。结论穿心莲内酯对脓毒症小鼠淋巴细胞免疫功能具有调节作用。 Objective To investigate the effects of andrographolide on the immune function of lymphocytes in mice with experimental sepsis. Methods Sepsis model was induced by cecal ligation and puncture (CLP). Forty eight C57BL/6 mice were randomly divided into 4 groups (n= 12) : sham group, CLP group, dimethyl -sulfoxide (DMSO) group and andrographolide (AND) group. Normal saline, DMSO and andrographolide were injected intraperitoneally in equal volume to each group respectively at 1, 6 h and 12 h after surgery. The spleen and thymus of mice in each group were harvested 24 h after operation. CD4+ T, CD8+ T, CD19+ B lymphocytes, and CD11c+ dendritic cells (DCs) were assessed using fluorescence activated cell sorter in spleen. Apoptosis of CD3+ T lymphocytes was detected using fluorescence activated cell sorter. Results The counts of CD4+ T, CD8+ T, CD19+ B lymphocytes and DCs in the AND group were (8.03±0.62)×105/ml, (3.42±0.58)×105/ml, (6.48±1.13)×106/ml and (5.50±0.79)×106/ml, higher than those in the CLP group (P〈0.01). Apoptosis of CD3+ T cell was inhibited in thymus in the AND group compared to that in the CLP group(P〈0.01 ). Conclusions Andrographolide could modulate the immune function of lymphocytes in mice with CLP sepsis.
