摘要
目的研究赤芍有效部位对KK/upj-Ay小鼠自发高血糖的影响,并对其化学成分进行分离鉴定。方法通过Sephadex LH-20富集赤芍有效部位,用KK/upj-Ay自发高血糖小鼠为动物模型,设低、中、高3个剂量组,罗格列酮为阳性对照药,C57BL/6J小鼠为空白对照组,观察有效部位对小鼠空腹血糖值的影响。同时用各种色谱技术对该部位化学成分进行分离,通过波谱分析等方法鉴定结构。结果给药3周后,有效部位中剂量组小鼠空腹血糖值低于模型组,两组比较有显著性差异(P<0.01);并从该部位分离得到9个化合物:芍药苷(Ⅰ)、氧化芍药苷(Ⅱ)、苯甲酸(Ⅲ)、没食子酸(Ⅳ)、没食子酸乙酯(Ⅴ)、1,2,3,4,6-五-没食子酰葡萄糖(Ⅵ)、柚皮素(Ⅶ)、二氢槲皮素(Ⅷ)。结论赤芍有效部位的降血糖功效可能与其具有改善胰岛素抵抗作用有关,化合物Ⅷ为首次从芍药属植物中分离得到。
Objective To study the effects of effective parts of Radix Paeoniae Rubra to KK/upj - Ay mice spontaneous hyper- glycemic and the chemical constituents. Methods Enrichment of the effective parts of Radix Paeoniae Rubra by Sephadex LH - 20, use KK/upj-Ay spontaneous hyperglycemia mice to do animal experiments, set low, medium and high three dose groups, rosigli- tazone for positive control, C57BL/rJ mice as blank control, in order to observe the effects of effective parts on fasting blood glu- cose levels in mice. And compounds were isolated by various column chromatography and their structurt were elucidated by spec- troscopic analysis. Results After 3 weeks, fasting blood - glucose of the mice in middle dose drug were less than that of the model group,with significant differences between the two groups (P 〈 0.01 ) ;Nine compounds were obtained from this part :Paeoniflorin ( I ), Oxypaeoniflora ( II ), Benzoic acid ( m ), Gallic acid ( IV ), Ethyl Gallic ( V ), 1,2,3,4,6 - pentagalloyl - [3 - glucose ( V[ ), Naringenin ( VII ), Dihydroquercetin ( VIK ). Conclusion The hypoglycemic effect of the effective parts of peony root may owe to it improve on insulin resistance, compounds VIII are obtained from the genus for the first time.
