摘要
以酿酒酵母(S.cerevisiae)细胞为正筛选模型的筛选结果显示,中草药黄连醇提液能挽救酿酒酵母在Calcofluor white(CFW)抑制浓度下的生长.利用酿酒酵母几丁质合成酶缺陷型菌株的最低抑菌浓度试验发现,几丁质合成酶Ⅲ缺陷型菌株(CHT003)对黄连醇提液的敏感性比野生型大4倍.几丁质含量测定结果表明:黄连醇提液处理后野生型酵母和缺陷型菌株CHT003的几丁质含量明显降低,由此推测黄连醇提物具有明显的抑制酵母细胞壁几丁质合成酶Ⅲ的活性成分.
Using yeast (S.cerevisiae) as positive screening model,the rescue effects of the extracts from Chinese medicinal herb on the growth of yeast strain from inhibition by Calcofluor white (CFW) was tested.The result showed that the extract of Coptis chinensis Franch could rescue the growth of yeast strain from the inhibition by CFW.The minimum inhibitory concentrations (MICs) of the extract on the wild strain WHU2a,mutant CHT020 (Chs1Chs3) and CHT003 (Chs1Chs2) was 2 600 μg/mL,1 300 μg/mL and 650 μg/mL,respectively.The chitin content of the wild strain and CHT003 were obviously decreased when treated with the extract.These data suggest that Coptis chinensis Franch extract has a obviously inhibitory effect on fungal chitin synthases Ⅲ.
