摘要
目的探讨CRIPTO-1在宫颈鳞癌组织中的表达及临床意义。方法采用免疫组化和原位杂交法检测90例宫颈鳞癌、30例宫颈上皮内瘤变及30例正常宫颈黏膜组织中CRIPTO-1蛋白及mRNA的表达情况,并分析其表达与宫颈鳞癌临床病理特征的关系。结果 CRIPTO-1蛋白在宫颈鳞癌组织中的阳性表达率(65.6%)高于宫颈上皮内瘤变组织(60.0%)及正常宫颈黏膜组织(10.0%),差异有统计学意义(χ2=28.571,P<0.001)。CRIPTO-1蛋白过表达与癌的浸润深度、淋巴结转移及TNM分期均关系密切(χ2分别为10.676、9.718及11.227;P均<0.05)。CRIPTO-1 mRNA在宫颈鳞癌组织中的阳性表达率(45.6%)高于宫颈上皮内瘤变组织(23.3%)及正常宫颈黏膜组织(10.0%),差异有统计学意义(χ2=14.577,P=0.001)。CRIPTO-1 mRNA过表达与癌的浸润深度、淋巴结转移及TNM分期均关系密切(χ2分别为13.741、4.720及30.170;P均<0.05)。CRIPTO-1蛋白及mRNA的表达呈正相关关系(r=0.287,P=0.006)。结论 CRIPTO-1的过表达可能与宫颈鳞癌的发生、发展有关,并对判断宫颈鳞癌患者的生物学行为有一定意义。
Objective To investigate the expression and clinical significance of CRIPTO-1 in cervical squamous cell carcinoma. Methods Immunohistochemical staining and in situ hybridization were used to detect CRIPTO-1 protein and mRNA expressions in the 90 cases of cervical squamous cell carcinoma, the 30 cases of cervical intraepi- thelial neoplasia, the 30 cases of normal cervical epithelia, and the relationship between CRIPTO-1 expression and clinicopathological features of cervical squamous cell carcinoma was analyzed. Results The positive expression rate of CRIPTO-1 protein in the cervical squamous cell carcinoma (65.6%) was higher than those in the cervical intra- epithelial neoplasia (60.0%) and the normal cervical epithelia (10.0%) , and there was a significant difference a- mong the three groups (χ2 =28. 571 ,P 〈 0. 001 ). The expression of CRIPTO-1 protein was closely correlated with the infilatration,lymphatic metastasis and TNM grade in the cervical squamous cell carcinoma (χ2 = 10. 676,9. 718 and 11. 227 ,P all 〈0.05 ). Conclusion The overexpressionof CRIPTO-1 may be associated with the carcinogenesis and development of cervical squamous cell carcinoma, and can be used for judging its biological behavior.
