摘要
目的 :研究参麦注射液对实验性心力衰竭 (心衰 )大鼠左室舒缩功能及血浆血管紧张素 (Ang )、内皮素 (ET)和心钠素 (ANP)的影响。方法 :采用缩窄腹主动脉方法复制大鼠心衰模型。左心室插管术测定血液动力学指标 ;放免法测定血浆 Ang 、ET和 ANP 浓度。结果 :心衰大鼠左室舒缩功能减退 ,血浆 Ang 、ET和 ANP水平显著升高 ;与假手术组相比 ,模型组大鼠血浆 Ang 、ET、ANP浓度分别升高 5 6 .2 1%、37.83%和44 .42 %。与模型组相比 ,参麦注射液高剂量组大鼠 Vmax、+ dp/ dtmax、 dp/ dtm ax、左心室内压峰值 (L VSP)分别升高 2 3.2 4%、10 .6 0 %、18.76 %和 17.31% ,左心室舒张末压 (L VEDP)降低 5 6 .15 % ,血浆 Ang 、ET、ANP浓度分别降低 32 .6 5 %、38.5 2 %和 9.44 % ,均有显著性差异 (P均 <0 .0 5 ) ;参麦注射液中、低剂量组大鼠各项指标均有不同程度的改善。结论 :参麦注射液能改善心衰大鼠左室舒缩功能 ,降低血浆 Ang 、ET浓度 ,影响 ANP分泌。
Objective:To investigate the effects of Shenmai injection (SMI,参麦注射液) on the plasma angiotensin(AngⅡ),endothelin(ET),atrial natriuretic polypeptide(ANP) and the myocardial contractility and relaxation of left ventricular in experimental rats with heart failure(HF).Methods:The wistar rat model with HF was produced by constricting abdominal aorta.Hemodynamic parameters,including maximum rate of intraventricular pressure rise(+dp/dtmax),left ventricular systolic pressure(LVSP),maximum velocity of contractile element shortening(Vmax),maximum rate of intraventricular pressure down(dp/dtmax) and left ventricular end diastolic pressure(LVEDP) were measured by the method of catherization.Plasma concentrations of ANP,ET and AngⅡ were determined by immunoradiometric assay.Results:In high dose SMI group,the LVSP,+dp/dtmax,dp/dtmax and Vmax were increased by 17 31%,10 60%,18 76% and 23 24%;LVEDP was decreased by 56 15%,plasma concentrations of AngⅡ,ET and ANP were decreased by 32 65%,38 52% and 9 44%,respectively.In comparison with those in model group,the differences were significant (all P <0 05).In middle dose group,the LVSP,+dp/dtmax, dp/dtmax and Vmax were increased by 14 76%,2 97%,14 19% and 11 40%,AngⅡ,ET and ANP were decreased by 22 47%,34.65% and 17 18%,respectively,the differences were significant in comparison with those in model group (all P <0 05).In low dose SMI group,the indices of hemodynamics were not considerable difference except LVEDP,but levels of plasma ANP,ET and AngⅡ were significantly lower than those in model group,decreased by 18 95%,33 59% and 16 77%,respectively.Conclusions:SMI can reduce the plasma concentrations of AngⅡ,ET and ANP,and improve the myocardial contractility and relaxation of left ventricular in heart failure rats.
