摘要
目的:观察60Coγ射线照射对大鼠腮腺和下颌下腺Mre11蛋白表达及细胞凋亡的影响。方法:取近交系Wistar成年大鼠60只,分为正常对照组(10只)及放射组(50只),放射组用60Coγ射线照射大鼠头颈部,总吸收剂量为15 Gy,每次照射3Gy,1次/d。照射3、6、9、12、15 Gy后2 h分别随机处死10只大鼠,用Western blotting法检测Mre11蛋白的表达;TUNEL法检测照射后细胞凋亡情况。结果:照射后唾液腺组织中Mre11蛋白的表达水平和细胞凋亡率均随放射剂量的增大而增高,Mre11蛋白的表达水平在腮腺照射9 Gy后和下颌下腺照射6 Gy后到达最大值,细胞凋亡率在腮腺照射9 Gy后和下颌下腺照射12Gy后到达最大值,之后开始逐渐减小,差异均具有统计学意义(P<0.05)。结论:治疗量60Coγ射线照射后大鼠唾液腺组织中Mre11蛋白的表达水平和细胞凋亡率与放射剂量及唾液腺类型有关。
Objective: To detect Mrell protein expression and cell apoptosis in rat salivary glands after 60 Coγ irradiation. Methods: 60 wistar rats were randomly divided into control group( n = 10) and 60 Coγ irradiation group (n = 50). 10 rats were respectively sacri- riced 2hs after irradiation of the dose of 3, 6, 9, 12 and 15 Gy respectively. Mrel I protein in salivary glands expression was assayed by western blotting, cell apoptosis was detected by TUNEL method. Results: In parotid gland after 60Co γ iraadation (Gy) at 0, 3, 6, 9 and 15 the relative expression of Mrel 1/β- actin was O. 15 ± O. 02, O. 23 ± O. 02, O. 35 ±O. 04, O. 54 ± O. 08, O. 36± O. 04 and O. 24 ± O. 03 (P 〈 O. 05 ) ; in submandibular gland O. 03 ± O. 02, O. 20 ±O. 04, O. 40 ± O. 05, O. 27 ± O. 04, O. 14± O. 04 and O. 09 ±O. O1 (P 〈 O. 05 ), respectively; the cell apoptosis ratio( % ) in parotid gland was O. 02 ± O. 00, O. 45±O. 05, O. 50 ± O. 06, O. 53 ± O. 04, O. 44 ±O. 03 and O: 41 ± O. 05 ( P 〈 O. 05 ) ; in submandibular gland O. 02± O. 01, O. 30 ± O. 02, O. 34 ±O. 01, O. 41 ± O. 06, O. 45 ± O. 04 and O. 29 ± O. 02 ( P 〈 O. 05 ) , respectively. Cortdusion: After 60Co γ irradiation, the expression of Mrel 1 protein and the cell apoptosis ra- tio in salivary glands were related to the radiation dose and the types of the salivary gland.
