摘要
目的:观察Smad2和Smad3在大鼠颅骨矢状缝牵张成骨过程中的时空表达。方法:建立大鼠颅骨矢状缝扩张培养模型,运用RT-PCR及免疫组织化学SABC技术,观察Smad2和Smad3 mRNA与蛋白在受力骨缝中的表达情况。结果:在牵张力的作用下,Smad2与Smad3的基因表达在20 min左右达到高水平,随后下降。骨缝受力后Smad2与Smad3蛋白表达显著增强,其阳性细胞主要集中在骨缝边缘区域。结论:Smad信号通路可能参与了骨缝细胞力学信号向生物化学信号的转化。
Objective: To observe the spatial and temporal expression of Smad2 and Smad3 in the stretched rat calvarial sagittal su- ture. Methods:By means of an organ cultured calvarial suture stretch model, RT-PCR and immunohistochemisty were used to detect the expression of Smad2 and Smad3 mRNA and protein in the stretched or non- stretched sutures. Results: Under the stimulation of dis- traction force, the mRNA of Smad2 and Smad3 reached the peak expression 20 min after stretch and then decreased. The immunostain- ing of Smad2 and Smad3 protein was increased and located in the osteoblasts adjacent to the edges of the sutures. Conclusion: Smad signal pathway may play a role in the process of the conversion from mechanical stress to biochemistry response.
