硫酸镁对脑缺血再灌注损伤保护作用的实验研究 被引量:3

摘要 目的 :探讨不同时间、不同剂量硫酸镁对大鼠局灶性脑缺血再灌注损伤后神经功能的保护作用。方法 :50只雄性SD大鼠被随机分为再灌注前高硫酸镁组、再灌注前低硫酸镁组、再灌注后高硫酸镁组、再灌注后低硫酸镁组和对照组共五组。采用改良的Longa’s法制作脑缺血再灌注模型 ,术后 4h予以再通。术后 6h采用胡氏脑组织微量分析法 ,测定梗死脑组织的Na+、K+、水含量 ,术后 2 4h先进行神经病学评分 ,再断头取脑染色后进行梗死灶体积的测定。结果 :硫酸镁治疗组与对照组相比 ,梗死灶体积缩小 ,脑水肿减轻 ,神经功能恢复好。治疗效果高硫酸镁组优于低硫酸镁组 ,再灌注前组又优于再灌注后组 ,其中再灌注前高硫酸镁组效果最为显著。结论 :硫酸镁对脑缺血后再灌注损伤有明显地神经保护作用 ,且治疗效果与治疗时间早晚和剂量大小密切相关。 Objective:To investigate the neuroprotective effects of MgSO 4 in different doses at varying stages after focal cerebral ischemia reperfusion injury.Methods:Fifty male SD rats were divided into four treatment groups,which including higher and lower doses of MgSO 4 before reperfusion group,higher and lower dose of MgSO 4 after reperfusion and one control group.Focal cerebral ischemia was carried out in rats with modified Longas model.Reperfusion occurred after 4 hours of ischemia.Hu's microanalysis was used to estimate sodium(Na +),potassium(K +)and water content in ischemia cerebral tissue at 6 hours after ischemia.Neurologic examinations were performed at 24 hours after ischemia,then infarct volume was measured with TTC staining. Results:Compared with control group,MgSO 4 can decrease infarct size,ameliorate brain edema and improve neueologic score.Animals given 125 mg/kg MgSO 4 is more effective than that given 62.5mg/kg MgSO 4.Neuroprotection is greater in rats given MgSO 4 prior to 1 hour of reperfusion than that in those posterior to 1 hour of reperfusion.Analysis of variance confirmed statistically significant effect of MgSO 4 treatment on infarct volume,brain edema and neurological score,especially in higher dose of MgSO 4 before reperfusion group.Conclusion:The neuroprotective effect of intraarterid MgSO 4 in this model is robust,dose dependent and related to the duration of ischemia.
