摘要
目的 建立聚合酶链反应快速检测加德纳菌方法 ,用于细菌性阴道病的诊断和治疗 .方法 根据加德纳菌基因序列中 IIS- 2 3sr RNA基因区设计并合成特异性引物 ,应用聚合酶链反应扩增阴道或宫颈分泌物中的加德纳菌 ,并与革兰染色和分离培养进行对照检测 .结果 经对阴道加德纳菌和 5种常见的阴道感染病原体检测 ,证实所用引物具有较高的特异性 ;敏感性实验结果 10 2 可扩增出明显的区带 ;在 5 0例宫颈分泌物标本中 ,革兰染色阳性检出率为 18% (9/ 5 0 )、分离培养阳性检出率为 16 % (8/ 5 0 ) ,聚合酶链反应阳性率为 2 8% (14/5 0 ) .结论 采用聚合酶链反应检测阴道分泌物中的加德纳菌 ,用于细菌性阴道病的快速诊断 。
AIM To establish PCR method to rapidly detect Gv and to apply it to diagnose and to treat bacterial vaginosis. METHODS A pair of specific primers were designed according to the IIS 23s rRNA gene of Gv and used PCR method to detect its specific gene fragment. Grams staining and bacterial culture were also used to detect its cells. RESULTS The primer was proved to be of hign specificity through detection of Gv and five other pathogens of viginosis; it can detect 100 cells sensitively. In 50 cervical secretion samples, positive detective rate of grams staining and PCR were 18%(9/50).16%(8/50) and 28% (14/50)respectively. CONCLUSION The PCR method can be used as a major laboratory method for Gv detection.
