摘要
目的:观察天参胶囊对2型糖尿病大鼠的脂糖紊乱的作用。方法:高糖高脂饲料配合腹腔注射链脲佐菌素(STZ)35mg/kg建立2型糖尿病大鼠模型。糖尿病大鼠按血糖值随机分为模型组、罗格列酮组(4mg/kg)、天参胶囊低(180mg/kg)、中(360mg/kg)、高剂量组(720mg/kg)。天参胶囊低、中、高剂量组分别灌胃不同剂量的天参胶囊水溶液,罗格列酮组灌胃罗格列酮,模型组与对照组灌胃等量自来水,连续给药30d后取血,测定各组大鼠血脂、血糖、糖耐量、胰岛素耐量的变化,并检查胰腺组织的病理变化。结果:天参胶囊高剂量(720mg/kg)可降低大鼠血清总胆固醇(TC)和甘油三酯(TG),显著降低游离脂肪酸(FFA),显著升高高密度脂蛋白(HDLC)、空腹胰岛素(FINS)水平和胰岛素敏感指数(ISI),改善胰腺细胞病变。结论:天参胶囊能改善实验性2型糖尿病大鼠的糖代谢和脂代谢紊乱,其作用机制可能与增加模型大鼠对胰岛素的敏感性有关。
Objective: To observe the anti-diabetic effect of Tianshen Capsule in type 2 diabetic meUitus (T2DM) rats. Method: Type 2 diabetic rat model was established by diet of high sucrose and high fat combined with intraperitoneal injection of streptozotocin (STZ). According to the blood glucose levels, the models were randomly divided into 5 groups: model control group, rosiglitazone group (4mg/kg), Tianshen capsule low dosage group (180mg/kg), Tianshen capsule middle dosage group (360mg/kg) and Tianshen capsule high dosage group (720mg/kg). The rats were administrated by gavage for 30 days. The influence of Tianshen capsule on diabetic rats was investigated, inclu- ding blood glucose, blood lipid, glucose tolerance, insulin tolerance, the pancreatic tissue was observed by immunohistochemisty. Results: High dosage of Tianshen capsule could lower total cholesterol (TC) and Ttriglyceride (TG) ( P 〈 0.05) as well as free fatty acid (FFA) ( P 〈 0.01 ), and significantly increase high density lipoprotein ( HDLC), fasting insulin (FINS) and insulin sensitivity index ( P 〈 0.01 ). Tianshen capsule could also reduce pancreatic cells pathological changes. Conclusion : 720mg/kg Tianshen capsule can improve glucose metabolism and lipid metabolism of experimental type 2 diabetic rats by improving glucose metabolism and increasing insulin sensitivity.
