应用分子标记辅助选择培育兼抗稻瘟病和白叶枯病的水稻恢复系 被引量:16

Breeding of Rice Restorer Lines with Resistance to Blast and Bacterial Blight by Marker Assisted Selection

摘要 本研究以抗稻瘟病水稻品系BL1为母本、6份抗白叶枯病育种材料(BB1-BB5,Bin0)为父本配组构建分离群体,针对抗稻瘟病基因Pi25、抗白叶枯病基因Xa4、Xa21、xa5和xal3以及育性恢复基因RfS和Rf4,通过分子标记辅助选择筛选出20份候选恢复系,携带恢复基因Rf3和Rf4、稻瘟病抗性基因Pi25以及2-4个白叶枯病抗性基因,且在检测的所有标记座位上均呈纯合型。20份候选恢复系经稻瘟病混合菌种、稻瘟病单菌种和白叶枯病混合菌种分别接种,有11个在3个试验中都表现中抗以上;20份候选恢复系与炳lA、原98A和内香85A配制组合,经F1田间观察和结实率考查,来自10个候选恢复系的19个组合表现良好,F1结实率的平均值为78.3%,变幅为71.1%~84.0%。最终筛选出4个兼抗稻瘟病和白叶枯病的恢复系,其中3份源于BL1/BB1,携带Rf3、Rf4、Pi25、Xa4、Xa21和xa5;1份源于BL1/BB5,携带Rf3、Rf4、Pi25、Xa4、Xa21和xa13。本研究同时以2类水稻病害和育性恢复能力为选育目标,不仅拓宽了分子标记辅助选择在水稻育种中的应用,而且针对育性恢复能力的选育与传统育种中育性恢复能力的鉴定相比,省时省力。 In this study, 20 rice candidate restorer lines harboring targeted alleles for blast and bacterial blight res- istance were selected through marker-assisted selection (M_AS) using segregating populations derived from crosses between the maternal parent BL1 (the donor of fertility restore gene Rf3 and the blast resistance gene Pi25) and 6 parental lines (BB1-BB5 and BB10, the donor of Rf3 and Rf4 genes and various bacterial blight resistance genes including Xa4, Xa21, xa5 and xal3). Among the 20 rice candidate restorer lines, 11 displayed a medium or high level of resistance to rice blast and bacterial blight under field and greenhouse conditions. Agronomic traits of hybrids derived from these lines were evaluated. The results showed that all 19 crosses derived from 10 candidate restorer lines displayed elite agronomic trait with the spikelet fertility ranging from 71.1%-84.0%. Based on the performance of disease resistance and other agronomic traits, 4 promising restorer lines were selected, Among them, 3 lines derived from the cross of BL1/BB1 and all of them possessed 6 targeted genes including Rf3, Rf4 Pi25, Xa4, Xa21 and xa5, while the other line derived from the cross of BL1/BB5 possessed another set of targeted genes including Rf3, Rf4, Pi25, Xa4, Xa21 and xal3. In the present study, hybrid rice restorer lines with multiple disease resistance were selected effectively and efficiently through MAS, The molecular markers developed in this study would facilitate the utilization of the targeted genes in future breeding programs.
