摘要
目的:探讨逍遥散降低雌性兔膝关节软骨细胞凋亡的可能作用机制。方法:40只新西兰兔通过手术造成膝关节失稳型模型,1周后随机分为中药组、西药组、混合组和对照组各10只,驱赶行走4周,分别给药8周。造模后12周末处理动物,免疫组化法检测兔膝关节软骨细胞凋亡线粒体通路相关基因的表达。结果:中药组、西药组和混合组Bcl-2表达均显著高于对照组;Bax、Caspase-3与细胞色素的C表达均显著低于对照组。结论:逍遥散可明显上调Bcl-2表达,下调Bax、Caspase-3与细胞色素的C的表达,可能是逍遥散降低兔膝关节软骨细胞凋亡的机制之一。
Objective: To explore the possible mechanism of Xiaoyao Powder on reducing female rabbit knee articular cartilage apoptosis cell. Method: 40 New Zealand rabbits were modeled through operation for knee joint instability, a week later, they were divided randomly into four groups: traditional Chinese medicine group, western medicine group, mixed group and control group, 10 in each group. Each rabbit was t'orced to walk for 4 weeks, then was given medicine or saline perfusion by oral gavage for 8 weeks. 12 weeks later, the animals were executed, and the knee cartilage tissue apoptosis gene expressions were detected by immunohistochemical method. Results: The apoptosis expression Bcl-2 of traditional Chinese medicine group, western medicine group and mixed group were higher than control group, and the expression of Bax, Caspase-3 and Cytochrome C were lower, and there was significant difference. There was no significant difference among traditional Chinese medicine group, western medicine group and mixed group. Conclusion: The Xiaoyao Powder could decrease knee articular cartilage gene apoptosis expression of Bax, Caspase-3 and Cytochrome C and increase expression of Bcl-2, and this might be a possible mechanism of Xiaoyao Powder in reducing knee articular cartilage apoptosis cells in rabbits.
