摘要 应用全细胞膜片钳技术研究 BDNF对培养海马神经元 NMDA受体的调控作用。结果发现 ,培养 1 8d的海马神经元 NMDA诱发电流小 ,BDNF可快速、可逆地增加 NMDA诱发电流 ,而培养 1 0 ,1 4d的海马神经元 NMDA诱发电流大 ,BDNF增强 NMDA诱发电流不明显。本文结果提示 BDNF对功能低下的海马神经元 BDNF(brain derived neurotrophic factor is known to play an important role in learning and memory. To elucidate underlying mechanisms, whole cell patch clamp recording was used to assess the effect of BDNF on the responses of cultured hippocampal neurons to the glutamate receptor agonist NMDA (N methy D asparatic acid). We found that peak amplitude of NMDA evoked currents in cultured hippocampal neurons at day 18 in vitro decreased significantly over that of NMDA currents at day 10 or 14. Interestingly, the NMDA evoked currents were greatly enhanced by BDNF (50 ng/ml) rapidly and reversibly in cultured neurons at day 18, but not at day 10 or 14. These results suggest that BDNF may rescue the down regulated NMDA receptor.
