摘要
对血清、胰酶以及染色时间等可能影响犬瘟热病毒蚀斑形成的条件进行了摸索。结果发现 ,对必须同步接毒的CDV小熊猫 (LP)株来讲 ,其形成蚀斑的最佳条件是 :同步接毒 10h后 ,加入血清含量为 2 %的无酚红的DMEM营养琼脂 ,第 5d时 ,加入 1/ 5体积的胰酶含量为 1/ 80 0 0的含中性红的DMEM营养琼脂染色 ,于第 6d时 ,便可形成蚀斑。挑选不同大小的蚀斑 ,克隆 3次后 ,获得了蚀斑直径为 0 .4～ 0 .6 ,0 .6～ 0 .8及 0 .7～ 0 .
The in fluence factors on plaque forming test of caninedistemper virus(CDV) LP strain,such as the concentration of bovineserum,pancriatic and different staining time,were determined.The Verocells infected with CDV LP strain were cultured in Eagle's medumcontaining 2% newborn bovine serum and 1% agarose gel.After 5 days,the staining agarose gel were added and the plaque in monolayer Verocells with LP strain were formed until the sixth day.The plaque(0.7 to 0.9,0.6 to 0.8 and 0.4 to 0.6 mm)virus es were isol ated by threesequential plaque selections.
