摘要
目的探讨玉米肽(CP)结合有氧运动对肥胖大鼠骨骼肌蛋白质代谢的影响。方法雄性SD大鼠150只,随机选取15只作为安静对照组(CON),给予普通饲料,其余135只给予高脂饲料建肥胖模型。8w后将40只肥胖大鼠随机分为肥胖对照组(DIO)、酪蛋白组(DCA)、玉米肽组(DCP)、运动组(DEX)和玉米肽运动组(DCE),每组8只。玉米肽组和玉米肽运动组给予玉米肽饲料,酪蛋白组给予酪蛋白饲料,其它各组给予普通饲料。运动组和玉米肽运动组进行有氧运动,干预4w后处死取材。检测血浆中血脂四项、胰岛素和瘦素水平以及骨骼肌中mTOR、p70s6k、rps6及其相应磷酸化蛋白的表达水平。结果干预4w后,与肥胖对照组相比,玉米肽运动组大鼠体重以及脂肪系数显著下降;比目鱼肌和跖肌肌肉指数显著增加;甘油三酯、胰岛素和瘦素水平显著下降;玉米肽运动组大鼠骨骼肌mTOR、p70s6k和rps6及其相应磷酸化蛋白表达显著增加。结论玉米肽结合有氧运动有效促进了肥胖大鼠骨骼肌蛋白质合成。其可能机制是玉米肽结合有氧运动激活了mTOR下游信号通路蛋白的表达,提高了骨骼肌蛋白质翻译效率。
Objective To explore the intervention of corn peptide (CP) with or without treadmill exercise on protein metabolism in skeletal muscle of obese rats. Methods Male Sprague-Dawley rats (n=135) were fed high-fat diet for 8w as a dietary- induced obesity model (DIO, n=40). Then, 40 rats were randomly divided into five groups: DIO control group, D10+casein group, DIO+CP group, DIO+ exercise group and DIO+CP+exercise group (n=8, each group). Other rats (n=lS) were fed balanced diet as normal group. The dietary protein content of casein group was equal to CP diet (200 g/kg) and exercise (Treadmill Ex) was performed 15 m/rain for 60 rain/d, 6 days a week. The rats were killed after 4w. At the same time, the rats of all groups were sacrificed. Plasma triglycerides (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), insulin and leptin were measured. Muscle P-mTOPUmTOR, P-p70s6k/p70s6k and P-rps6/rps6 expressions were measured by Western-blotting. Results After 4w, compared with DIO control group, body weight and perirenal fat mass index in DIO+CP+exercise group were significantly decreased (P〈0.05) but soleus mass index was apparently increased (P〈0.05). TG, insulin and leptin in DIO+CP+exercise group were markedly decreased (P 〈0.05). But P-roTOR/ mTOR, P-p70s6k/p70s6k and P-rps6/rps6 expression of muscle were all obviously increased (P〈0.05). Conclusion Corn peptides consumption with exercise is able to promote protein synthesis of skeletal muscle in obese rats possibly by activating roTOR signaling pathway.
