Research Advance of Gene Knockout Technology and Its Application

摘要 基因敲除技术是基因功能研究的最直接和有效的方法之一。近年来发展起来多种基因组功能性研究的高效工具,包括ZFNs、TALENs和Cas9等,为高效率基因打靶开辟了一条全新的思路。这些基因编辑的革命性技术,能大力推动生物学、医学研究的发展。但对这些新技术还没有进行系统研究。文中对基因敲除发展历程、技术原理以及新技术的应用等进行概述,为基因敲除技术的进一步发展应用提供依据。 The gene knockout technology was deemed to be one of directly and effectively methods in gene function research. Recently, several effective tools, including ZFNs, TALENs, Cas9, were developed quickly and opened the new mentality for gene targeting. These revolutionary technologies of gene editing would significantly promote the development of biology and medicine. However, reviews for the new technology are still few. In this paper, it gives a quick overview of develop history, technical principle, application for gene knockout technology, which will nrovide a reference for fiJrther develonment and annlicatian of ~ene knocknm technaln^v.
