摘要
目的合成非天然氨基酸(S)-2,6-二甲基苯丙氨酸。方法 2,6-二甲基苯甲酸经还原得到2,6-二甲基苯甲醇,再溴化生成2,6-二甲基溴苄,2,6-二甲基溴苄与(S)-BPB-Ni-Gly络合物缩合,经分离后得到(S)-Tmp-BPB-Ni络合物,经酸解后得到(S)-2,6-二甲基苯丙氨酸。结果以33.6%的总收率得到目标产物(S)-2,6-二甲基苯丙氨酸。结论该方法具有原料廉价、易得、反应条件温和等优点。
Objective To synthesize the unnatural amino acid (S)-2,6-dimethylphenylalanine.Methods 2,6-Dimethylbenzoic acid was reduced to produce 2,6-dimethylbenzyl alcohol,then the alcohol was brominated to generate 2,6-dimethylbenzy bromide.The bromide was then coupled with (S)-BPB-Ni-Gly complex to give (S)-Dmp-BPB-Ni complex,which was then decomposed by acid to obtain (S)-2,6-dimethylphenylalanine.Results (S)-2,6-dimethylphenylalanine was synthesized at a total yield of 33.6%.Con-clusion This method has characteristics of cheap starting material and simple experimental procedure.
