中性蛋白酶高产菌株的筛选及产酶条件的优化 被引量:1

The screening and optimization of fermentation conditions of a high-producing neutral protease strain

摘要 从土壤中筛选出6株中性蛋白酶菌株,并对其中酶活力较强的1株枯草芽孢杆菌的发酵产酶条件进行了优化。最终得到最佳培养基配方为:玉米粉1%、硫酸铵0.6%、麸皮3%、Na2HPO4·12H2O 0.4%,KH2PO40.03%,CaCl20.1%,pH 6.0。最适培养条件为:发酵时间48 h、发酵温度30℃,酶活力最高可达1 475.6 U/mL。 6 neutral protease strains were screened from the soil, among which a highest enzyme produ- cing strain bacillus subtilis was studied on its optimum neutral protease producing conditions by fermenta- tion. The results showed that the optimum culture medium was prepared with corn flour of 1%, (NH4) 2SO4 of 0.6% ,bran of 3% N a2HPO4 12H2O of 0.4% , KH2PO4 of 0.03% , CaC12 of 0.1% , pH of 6.0. And the optimum neutral protease producing conditions were as follows:fermentation time 48 h, fermentation temperature 30 ℃. In these conditions, the highest enzyme activity reached 1 475.6 U/mL.
