摘要
建立HPLC-ELSD检测岩藻聚糖硫酸酯中岩藻糖含量的方法。通过HPLC-ELSD方法检测,色谱条件为:Prevail Carbohydrate ES分析柱(5μm,4.6×250 mm),柱温:30℃,蒸发光散射检测器,漂移管温度:100℃,气流:3 L/min,流动相:80%乙腈-0.5%醋酸胺(90∶10)。结果表明,岩藻糖峰面积对数与浓度对数成良好线性关系,其线形范围为276.3μg/mL^2 210.4μg/mL,r=0.999 8(n=5)。回收率:99.97%,RSD:1.49%(n=9)。该法可准确检测岩藻聚糖硫酸酯中岩藻糖含量,各成分可良好分离,结果重现性好。
To establish a HPLC-ELSD method for determining L (-) fucose in fucoidan extract. HPLC condition: Prevail Carbohydrate ES analytic column (5 μm ,4.6×250 mm), Column Temperature: 30℃, ELSD drift tube: 100℃ ; Air Flow : 3 L/min, Mobile Phase : 80 % acetonitrile -0.5 % ammonium acetate (90 : 10 ). Linear range of L (-) fucose logarithm equation is 276.3 μg/mL-2 210.4μg/mL, (r=0.999 8, n=5). Recovery: 99.97 %. RSD: 1 A9 % (n=9). This method can detect L (-) fucose in focoidan extract accurately, every component effective segregation, result have good repeatability.
