人脐带间充质干细胞与骨髓间充质干细胞分化为胰岛素分泌细胞的能力比较 被引量:6

摘要 目的:比较人脐带间充质干细胞(UC-MSCs)与骨髓间充质干细胞(BM-MSCs)分化为胰岛素分泌细胞的能力。方法:分离获得UC-MSCs与BM-MSCs,用10 mmol/L尼克酰胺,2 nmol/L活化素A,10 nmol/L唾液素4,100 pmol/L肝细胞生长因子,10 nmol/L胃泌素对两组细胞进行诱导分化,从基因表达、蛋白表达及激素分泌水平上比较其分化能力。结果:与BM-MSCs分化组相比,UC-MSCs分化组胰岛素分泌能力、胰十二指肠同源盒1、C-肽及胰高血糖素表达水平及分化后细胞活力更强。结论:UC-MSCs比BM-MSCs具有更强的胰岛素分泌细胞分化能力,结合其来源稳定及免疫原性低,可以作为干细胞治疗糖尿病的理想细胞来源。
