索法酮在幽门螺杆菌感染的胃上皮细胞中的体外抗炎效应 被引量:1

摘要 目的:探讨索法酮(sofalcone,SFN)对幽门螺杆菌(Helicobacter pylori,Hp)感染的胃上皮细胞的体外抗炎效应。方法:体外培养胃上皮细胞系AGS,加入SFN和培养的Hp感染不同时间。Western blot检测AGS细胞中IκBα,环氧化酶2(cyclooxygenase-2,COX-2)蛋白表达情况;实时定量PCR检测COX-2和MUC-2mRNA表达;荧光探针和ELISA分别检测ROS的和IL-8的产生。结果 :SFN处理后,IκBα表达显著增加,且能抑制NF-κB活化,并降低COX-2、ROS和IL-8表达水平。同时也能增加黏蛋白MUC-2 mRNA表达。结论:SFN能抑制了NF-κB的活化以及相关炎症因子的产生,并能增加黏蛋白的表达显示出强大的预防Hp感染的胃上皮细胞炎症潜能。 Objective To investigate the anti-inflammatory effect of sofalcone (SFN) in H. pylori-infected gastric epithelial cells in vitro. Methods Gastric epithelial cell line AGS was cultured in vitro, and then was incubated with SFN and H. pylori for different lengths of time. Expressions of IKBα and cyclooxygenase-2 (COX-2) were detected by Western blot; the mRNA levels of COX-2 and MUC-2 were measured by real-time PCR; and production of reactive oxygen species (ROS) and IL-8 was detected by fluorescent probe and ELISA, respectively. Results After treatment with SFN, IKBα expression was significantly increased, nuclear factor kappa B activation was inhibited, and expressions of COX-2, ROS, and IL-8 were decreased. Meanwhile, SFN also increased the mRNA expression of MUC-2. Conclusions Sofalcone effectively inhibits activation of NF-KB and production of related inflammatory factors, while increases MUC-2 expression in H. pylori-infected AGS cells. It shows great potential as a candidate agent for the prevention of H. pylori-induced inflammation in gastric epithelial ceils.
