摘要
目的:探讨温阳振衰颗粒对慢性充血性心力衰竭(CHF)模型兔心脏功能和内皮素-1(ET-1)的影响。方法:将43只兔随机抽取11只为正常对照组,利余32只耳缘静脉注射阿霉素建立CHF模型,连续8周。4周后对其行心脏彩超检查,并对存活的30只造模实验兔根据心功能分层随机分为模型8周组,依那普利组,温阳低、中、高剂量组,每组6只,第5周开始,各治疗组分别灌胃给药,正常对照组及模型8周组均以等量生理盐水灌胃,每天1次,连续4周。灌胃结束后观察比较各组模型兔心脏功能及ET-1的变化。结果:与模型8周组比较,各治疗组均可改善模型兔LVEF、LVFS、E/A(P<0.05),并均能不同程度降低ET-1(P<0.01),且温阳高剂量组疗效优于中、低剂量组。结论:温阳振衰颗粒可通过调节心衰模型兔心脏功能及降低血浆中ET-1的含量以防止和延缓心衰的进一步恶化,且高剂量组疗效优于中、低剂量组。
Objective：To investigate the effects of Wenyang Zhenshuai granules （for warming yang and invigorating weakness） on cardiac function and endothelin - 1 （ ET - 1 ） level in a rabbit model of chronic congestive heart failure （CHF）. Methods ：Forty - three rabbits were randomly divided into normal control group （ n = 11 ） and CHF model group （ n = 32 ）. In the CHF model group, adriamycin was intravenously injected into the ear margin for 8 weeks to establish a CHF model. Four weeks later,the treated rabbits underwent echocardiography, and the 30 surviving rabbits were randomly and equally divided into 5 groups according to cardiac function：8 week model group, enalapril group, and low - , middle - , and high - dose Wenyang Zhenshuai groups. Since the 5th week, the treatment groups received respective treatments by garage once daily for 4 weeks, and the normal control group and 8 week model group received an equal volume of normal saline once daily for 4 weeks. These groups were compared with respect to changes in cardiac function and ET - 1 level. Resuhs：Compared with the 8 week model group, all treatment groups showed improvements in left ventricular ejection fraction ,left ventricular fractional shortening,and E/A ratio （ P 〈 0. 05 ） and variable decreases in ET - 1 level （ P 〈 0. 01 ）. The high - dose Wenyang Zhenshuai group had a significantly better outcome than the middle - and low - dose Wenyang Zhenshuai groups. Conclusion：For the rabbit model of CHF ,Wenyang Zhenshuai granules can delay the progression of CHF by improving cardiac function and reducing plasma ET - 1 , and high - dose Wenyang Zhenshuai granules have better efficacy than middle - and low - dose Wenyang Zhenshuai granules.
