摘要
目的 :观察葡萄球菌肠毒素 A的细胞增殖诱导能力及其抗肿瘤作用。方法 :MTT微量酶反应比色法。结果 :纯化 SEA在 1 0 - 12 ~ 1 0 - 5g/ml浓度范围对体外培养的 BALB/C鼠脾细胞表现了细胞增殖诱导能力 ,并呈剂量依赖关系 ,其中 1 0 - 7~ 1 0 - 5g/ml SEA的作用强于最适量 (2 5μg/ml) PHA。在 E/T为 5~ 2 0∶ 1条件下 ,1 0 - 5g/ml SEA活化 48小时的 BALB/C鼠脾细胞对 Yac- 1细胞的杀伤活性高于 NK细胞 ,但 SEA未能增强 BALB/C鼠脾细胞对 B16细胞的杀伤活性。结论 :葡萄球菌肠毒素 A具有较强的细胞增殖诱导能力 。
Objective:To observe the cellular grow induced capability and antineplastic of staphylococcal enterotoxin A.Methods:In method with MTT.Results:The Purified SEA showed an increasing capability on BALA/C of spleenocyte in mice at dosage of 10 -12 ~10 -5 g/ml concentrations in vitro and in dose dependent manner.The SEA at 10 -7 ~10 -5 g/ml had more effect than PHA at optimum dose (25μg/ml).When E/T was at 5~20∶1,the activated spleenocyte BALB/C of mice by 10 -5 g/ml SEA for 48hr had more effect than NK in killing Yac 1 cell,but no marked increase in B 16 cell.Conclusion:Staphylococcal enterotoxin A has the better cellular grow induced capability,so that the capability of superantigen might be used in the anti tumor biological therapeutically rfficiency.
