青海地区五脉绿绒蒿总黄酮含量测定参数及测定条件设定 被引量:1

SETTING THE PARAMETERS AND CONDITIONS FOR DETERMINATION OF TOTAL FLAVONOID OF Meconopsis quintuplinervia IN QINGHAI

摘要 目的通过对青海地区五脉绿绒蒿总黄酮含量测定参数及测定条件的比较研究,确立适用于高海拔试验条件下绿绒蒿属药材中总黄酮含量的测定参数。方法采用单因素考察法对样品提取溶剂、水解酸液的浓度、时间进行了考察,对HPLC法测定五脉绿绒蒿药材中槲皮素、山奈素、异鼠李素的总含量的色谱系统进行了比较研究。结果 70%甲醇为提取溶剂提取率较高,盐酸-70%甲醇(5:25)水解1 h时酸水解率高。槲皮素、山奈素、异鼠李素3种成分进样量分别在0.026~0.26μg、0.0177~0.177μg、0.0053~0.053μg范围内呈良好的线性关系,相关系数均为0.9999。平均加样回收率(n=9)分别为97.61%、97.37%、97.34%。测得青海产5批五脉绿绒蒿总黄酮含量在0.513~1.271 mg﹒g-1之间。结论本研究确定的总黄酮的含量测定参数及测定条件能够准确测定绿绒蒿属药材中总黄酮含量,与原标准相比能够更好地反映高原地区绿绒蒿药材的整体质量。 Objective By comparing the parameters and conditions of the determination of total flavonoid of Meconopsis quintuplinervia in Qinghai,set the parameters for determination of total flavonoid of medicinal herbs of MeconoPsis which is applied in high altitude laboratory.Methods Investigated the extraction solvent,content of hydrolysis acid and reaction time with single factor exploration.Comparative study was carried on chromatographic system for determination of total content of quercetin,kaempferide,isorhamnetin of Meconopsis quintuplinervia by HPLC.Results The extraction rate of 70% methanol was higher,and the hydrolysis rate of HCL-70% methanol(5:25)was higher as well.Good linearity was obtained for quercetin,kaempferide and isorhamnetin with injection quantity 0.026 ~ 0.26 μg,0.0177 ~ 0.177 μg and 0.0053 ~ 0.053 μg,respectively,with a correlation coefficient R = 0.9999.The average spiked recovery is 97.61%,97.37%,97.34%,respectively.Total flavonoids of 5 batches of Meconopsis quintuplinervia in Qinghai ranged from0.513 mg/g to 1.271 mg/g.Conclusion The determination parameters and conditions setting in this paper is able to accurately determine the total flavonoid of MeconoPsis.Compared with previous specification,it can better reflect the overall quality of medicinal herbs of MeconoPsis in plateau.
