阿维A对寻常性银屑病患者树突状细胞的影响

Influence of Acitretin on Dendritic Cells in Patients with Psoriasis

摘要 目的研究阿维A调控银屑病患者外周血树突状细胞功能,探讨阿维A治疗银屑病作用机制。方法抽取银屑病患者外周血,分离单个核细胞,体外诱导培养树交状细胞(Dendritic cells,DCs)。不同浓度阿维A(Inmol/L和10nmol/L)作用DCs 24h,对照组为只加培养基,流式细胞术(Flow cytometry,FCM)检测DCs中CD80,CD83及CD86表达水平,酶联免疫吸附试验(ELISA)方法检测细胞培养上清液中IL-12表达水平。结果体外成功培养出DCs,流式细胞术检测到1,10nmol/L阿维A作用DCs24h后,CD80,CD83及CD86表达率分别为(69.74±3.83)%、(61.12±3.03)%、(70.18±3.32)%和(60.19±3.97)%、(50.78±4.81)%、(48.96±3.18)%,显著低于对照组DCs上CD80,CD83及CD86表达率(75.15±2.76)%、(72.43±1.94)%、(77.81±2.46)%(P<0.01)。1,10nmol/L阿维A作用DCs 24h后,DCs分泌到细胞培养液中的IL-12表达水平分别为(260.36±15.26)(226.98±17.24)显著低于对照组(282.80±23.26)(P<0.01)。结论阿维A可以降低银屑病患者外周血中DCs上CD80.CD83及CD86表达率,抑制DCs活化,且降低IL-12因子的分泌量,能够调控患者机体的免疫功能,从而对银屑病起到治疗作用。 Objective To study the regulatory role of acitretin on peripheral blood dendritic cell function in patients with psoriasis in order to explore the mechanism of acitretin in the treatment of psoriasis. Methods Mononu- clear cells were isolated from peripheral blood from psoriatic patients. Dendritic cells(DCs) were treated with lnmol/L and 10nmol/L acitretin for 24h. DCs treated with medium alone were served as controls. The expression levels of CD80, CD83 and CD86 on DCs were detected by flow cytometry(FCM) and the IL-12 levels in culture supernatant were detected by ELISA. Results In comparison with vehicle treatment, both 1 nmol/L and 10nmol/L acitretin significantly decreased the expression levels of CD80 [ (69.74± 3.83 )% for lnmol/L, (60. 19±3.97)% for 10nmol/L, vs. (75.15±2.76) % for vehicle], CD83[ (61.12 ±3.03% for lnmol/L, (50.78±4.81)% for 10nmol/L, vs. (72.43±1.94)% for vehicle] and CD86[(70.18±3.32)% for lnmol/L, (48.96±3.18)% for 10nmol/L, vs. (77.81±2.46)% for vehicle] (P〈0.01 for all). ELISA result showed that the IL-12 expression levels were siguificantly lower in acitretin-treated groups than in control group(260.36±15.26 for 1nmol/L and 226.98±17.24 for 10nmol/L, vs. 282.80±23.26 for control) (P 〈 0.01 ). Conclusion Acitretin can reduce the expression of CD80, CD83 and CD86, inhibit the activation of DCs, and decrease the secretion of IL-12. The therapeutic effect of acitretin on psoriasis could be via regulation of patients' immune function.
