基于磷含量调控的红霉素发酵培养基优化

Optimization of Fermentation Medium for Erythromycin by Regulation of Phosphate Content

摘要 本研究依据有机氮源磷含量不同设计了3种培养基,试验结果表明,低磷培养基(黄豆饼粉2.0%、脱脂豆粉1.0%和羽毛蛋白胨1.0%)条件下红霉素效价较高,为11 209?g/ml。为了降低发酵后期黄豆饼粉中磷的释放所造成的红霉素合成受阻遏,在上述培养基的基础上用0.5%硫酸铵和0.031%磷酸二氢钾替换1.0%黄豆饼粉(替换前后氮磷总量基本相等),红霉素效价达11 657?g/ml,同时发酵液后期黏度约降低20%。 Three fermentation media with different phosphate contents were designed. The highest erythromycin titer of 11 209 μg/ml was obtained under the low level phosphate medium containing 2.0% soybean meal, 1.0% defatted soybean powder and 1.0% feather peptone. In order to avoid the repression of erythromycin biosynthesis by phosphate released from the soybean meal, 1.0% soybean meal was substituted by 0.5% ammonium sulfate and 0.031% potassium dihydrogen phosphate. The erythromycin production reached to 11 657 μg/ml. The broth viscosity decreased by 20%.
