Synthesis of Dexlansoprazole

摘要 用2-氯甲基-3-甲基-4-(2,2,2-三氟乙氧基)吡啶盐酸盐和2-巯基苯并咪唑经缩合制得2-[3-甲基-4-(2,2,2-三氟乙氧基)吡啶-2-基]甲巯基-1H-苯并咪唑,再经过氧化氢二异丙苯不对称氧化和乙醇重结晶"一锅法"合成(R)-兰索拉唑,收率75%,ee值99.9%。 Dexlansoprazole was synthesized from 2-chloromethyl-3-methyl-4-(2,2,2-trifluoroethoxy)pyridinehydrochloride and 2-mercaptobenzimidazole by condensation to give 2-[3-methyl-4-(2,2,2-trifluoroethoxy)pyridin-2-yl]methylthio-1H-benzimidazole, followed by asymmetric oxidation and purification with the yield of 75% and optical purity of 99.9%.
