去氢茯苓酸在大鼠体外肝微粒体中的生物转化研究

STUDIES ON THE BIOTRANSFORMATION OF DEHYDROPACHYMIC ACID IN RAT LIVER MICROSOMES IN VITRO

摘要 本文主要研究蒙药材茯苓中主要化学成分之一的去氢茯苓酸在大鼠肝微粒体中的生物转化。用大鼠肝微粒体体外温孵法进行去氢茯苓酸的生物转化,高效液相色谱法检测和制备原形化合物去氢茯苓酸及其生物转化产物,核磁共振波谱法和质谱法确定生物转化产物的结构。优化了温孵体系;去氢茯苓酸在苯巴比妥诱导的大鼠肝微粒体药物代谢酶作用下,产生2个转化产物,即茯苓酸和25-羟基茯苓酸。去氢茯苓酸可被苯巴比妥诱导的大鼠肝微粒体药物代谢酶转化为茯苓酸和25-羟基茯苓酸。 To study the biotransformation of dehydro pachymic acid, which is one of main chemical constituents in the sclerotium of Poria cocos ( Schw. ) Wolf as a traditional Chinese medicines, using in vitro drug - metabolized enzymes prepared from the phenobarbi- tal - induced rat liver microsomes. After incubation of dehydropachymic acid with drug - metabolized enzymes prepared from the phe- nobarbital - induced rat liver microsomes at 37 ? C in vitro, the high - performance liquid chromatography method was applied to ana- lyze and prepare original dehydropachymic acid and its biotransformated products. The their chemical structures were determinated by NMR and MS techniques and compared with those of authentic compounds. The constitution of the incubation system was optimized. Dehydro acid could be transformed by drug -metabolized enzymes in rat liver microsomes to produce two the biotransformated products pachymic acid and 25 - Hydroxy pachymic acid. Dehydro - pachymic acid could be transformed by drug - metabolized enzymes in rat liver microsomes to pachvmic acid and 25 - Hvdroxv pachvmic acid.
