左乙拉西坦对戊四氮激发癫痫大鼠海马区Panx1表达的影响 被引量:1

Influence of levetiracetam on Panx 1expression in the hippocampus of pentylenetetrazol-kindled rats

摘要 目的研究戊四氮(PTZ)激发癫痫大鼠海马区Panx 1表达的变化及左乙拉西坦(LEV)对其表达的影响。方法将32只SD大鼠随机均分为正常对照(A)组、癫痫模型对照(B)组、LEV 150mg/kg干预(C)组和甘珀酸(CBX)20mg/kg干预(D)组。B、C、D组采用PTZ腹腔注射制作癫痫大鼠模型。LEV和CBX干预,每天1次,持续2周。用Racine评分判断癫痫的发作情况,分别采用RT-PCR和Western blot检测各组大鼠海马区Panx 1的表达。结果与A组比较,B、C和D组的大鼠海马区Panx 1表达水平升高(P<0.05),与B组比较,C组和D组大鼠癫痫的发作减轻(P<0.01),海马区Panx 1的表达下降(P<0.05),且C组优于D组(P<0.05)。结论 PTZ致痫大鼠海马区Panx 1表达增多,LEV能抑制Panx 1的表达,减少模型大鼠癫痫的发作频率。 Objective To explore the influence of levetiracetam(LEV) on Panx 1 expression in the hippocampus of pentylenetetrazol(PTZ)-kindled rats. Methods Thirty-two SD rats were egually randomized into four groups of A(blank control), B(PTZ model control), C(treated with LEV 150 mg/kg) and D[-treated with carbenoxolone (CBX) 20 mg/kg]. The interventions of LEV and CBX were performed once a day for 2 weeks. The epileptic rat model in groups of B, C and D was established using PTZ intraperitonial injection. Epileptic seizures were evaluated by Racine score. The expression of Panx 1 in the hippocampus was detected by RT-PCR and Western blot. Results Compared with group A,Panx 1 expression was higher in groups of B,C and D(P^0. 05). Compared with group B, the attack of seizures was attenuated in groups of C and D(P%0. 01) with reduced Panx 1 expression(P〈0. 05), which was less in group C than that in group D (P^O. 05). Conclusion The expression of Panx 1 in the hippocampus is increased in PTZ-kindled rats, which can be suppressed by LEV with less attack of epileptic seizures.
