单链构象多态性分析技术检测结核杆菌利福平耐药基因 被引量:1

摘要 目的 :研究本地区临床分离结核杆菌rpoB基因突变与利福平 (RFP)耐药性的关系。 方法 :对 3 6株RFP敏感株和 44株RFP耐药株rpoB基因的 3 2 8bp的PCR扩增产物进行单链构象多态性 (Single strandcomformationpoly morphism ,SSCP)分析。 结果 :3 6株RFP敏感株的SSCP带谱均与参考株H 3 7Rv相同 ,44株RFP耐药株中 ,2 5株高度RFP耐药株和 11株低度RFP耐药株的SSCP带谱与H 3 7Rv带谱有差异 ;另 8株低度RFP耐药株的SSCP带谱与H3 7Rv带谱相同。结论 :SSCP分析可检测出rpoB基因突变 ,该基因突变与本地区临床分离结核杆菌对RFP耐药有关。 Objective:For the sake of studying the relationship of rpoB gene point mutation and rifampin resistance.Methods:The 328bp in rpoB gene of 36 RFP susceptible strains and 44 RFP resistant strains were amplified by PCR and investigated by single strand conformation polymorphism analysis(SSCP) with silver stain.Result:H37Rv was used as a control,36 RFP susceptible strains and 8 low level resistance to RFP strains showed same SSCP pattern,however,there is a difference in the SSCP pattern of 25 high level resistance to RFP and 11 low level resistance to RFP strains.Conclusion:SSCP technique could detect rpoB gene point mutation,which might related to RFP resistant strains.
