成骨细胞促进胚胎干细胞向造血前体细胞分化的研究

摘要 目的 观察成骨细胞对胚胎干细胞(ESC)向造血前体细胞分化的促进作用.方法 用平皿黏附法培养BALB/c小鼠骨髓基质细胞,诱导分化为成骨细胞,鉴定后用作支持细胞.将小鼠胚胎于细胞(ESC-D3)先诱导生成胚胎体(EB),再与成骨细胞共培养,3d后进行代表血液血管干细胞的原始细胞集落(BL-CFC)培养并计数;7d后行流式细胞仪检测ESC来源的CD34+细胞生成比例.结果 在成骨细胞的支持下,与空白对照比较,ESC来源的代表血液血管干细胞的BL-CFC集落增加了2.89倍;由ESC生成的CD34+细胞由(12.68±2.39)%增加到(32.83±3.84)%.结论 成骨细胞体外可促进胚胎干细胞向造血前体细胞分化. Objective To explore the role of osteoblasts in promoting the differentiation of embryonic stemcells (ESCs) into hematopoietic precursor cells.Methods The cultured bone marrow BALB/c mouse stromal cells using plating adhesion method were induced to differentiate into the osteoblasts which were identified and used as the supporting cells of subsequent experiments.The murine ESCs-D3 were induced to become the embryonic body (EB),then co-cultured with the osteoblasts.Three days later,the blast colony-forming cells (BL-CFCs) representing hemangioblasts cultures was done.Seven days later,the ESCs-derived CD34 + cells were detected by using flow cytometry.Results Supported by osteoblasts,as compared with the blank control,the number of ESCs-derived BL-CFCs (hemangioblasts) was increased by 2.89 times and the ratio of ESCs-derived CD34+ cells was increased from (12.68 ± 2.39)% to (32.83 ± 3.84) %.Conclusion Osteoblasts could promote the differentiation of ESCs into hematopoietic precursor cells in vitro.
