摘要
以粘质沙雷氏茵(Serratia marcescens)ATCC 31026为出发菌株,经过硫酸二乙酯(DES)、亚硝基胍(NTG)和紫外(UV)逐级诱变,选育出了一株具有3-氨基-1,2,4-三氮唑抗性(AMT^r)、6-巯基嘌呤抗性(6-MP^r)、组氨酸甲酯抗性(HE^r)、2-硫尿嘧啶抗性(2-TU^r)、D-组氨酸抗性(DH^r)等遗传标记的突变株ZJZ 625(AMT6-MP^rHE^r2-TU^rDH^r),L-组氨酸产量由最初的2.0 g/L提高到7.6g/L。对ZJZ 625进行培养基及培养条件优化,研究了单因素对发酵的影响,由响应面分析得出最佳培养基配方,使最终产量达到9.7 g/L。
The Serratia marcescens ATCC 31026 was used as the starting strain. By stepwise mutagenic treatments with N- methyl-N-nitro-N-nitrosoguanidine(NTG), diethylsulfate ( DES ) and ultraviolet ( UV ), the objective mutant ZJZ 625 ( AMTr6-MPrHEr2-TUr6-MEPrDHr) was obtained. The mutants was selected on the agar plates containing 3-amino-1,2,4 triazole, 6-mercaptopurine arabinoside, L-histidine methyl ester dihydrochloride, 2-thiouracil and D-histidine. The L-his- tidine producing mutant ZJZ 625 could accumulate 7.6 g/L. Based on the effects of single factors and then the response surface analysis was used to optimize the concentrations of the substrate. The amount of L-histidine accumulation reached to a level of 9.7 g/L.
