摘要
细胞凋亡与细胞坏死在形态学方面有着显著的差别 ,通常电镜观察结果是证明细胞凋亡的最可靠的证据。我们认为细胞凋亡的半薄切片甲苯胺蓝染色更加简便、快速 ,且较准确。本实验采用地塞米松诱导 Jurkat细胞凋亡的模型 ,采用半薄切片的光镜观察及超薄切片透射电镜观察。结果表明 ,半薄切片可以初步判断凋亡细胞 ,透射电镜观察是确定细胞凋亡的直接证据 ,且方法稳定 。
Observation: Apoptosis was usually proved by elec tron microscopy. The purpose of the study was to compare the effect of semithin section by microscope and ultrathin section by electron microscope in observation of apoptosis. Methodology: Apoptosis of Jurkat cell was induced by dexamethasone, a nd prepared to s emithin and ultrathin sections as routine. The 1μm survey semithin section was s tained with 1% toluidine blue solution. Results: Condensation of nuclear chromat in, dense granular nuclear fragment and formation of apoptotic body was clearly distinguished in semithin section 24h after dexamethasone treatment, which was a lso observed in ultrathin section. Conclusion: Semithin section was stable and believable to detect apoptosis as ultrathin section.
