摘要
目的:建立三碘甲腺原氨酸(T3)第三次定量免疫测定用国家标准品。方法和结果:精确称取高纯T3原料,用含0.5%BSA的0.02M PBS缓冲液配制成100ng/ml溶液,经无菌过滤处理后,分装、冻干制成候选品。以T3第二次国家标准品(150550-0004)为对照品,经协作标定,候选品中T3免疫活性为50ng/支。结论:候选品各项指标均符合要求,可作为T3第三次免疫测定用国家标准品使用。本标准品的建立,为T3免疫测定用试剂盒的标准化提供定量基础。
Objective To establish the 3rd of national standardized products for quantitative immunoassay of 3, 5,3' -Triiodothyrenine(T3 )in China. Method and Result High purity of T3 raw material were precisely weighted to prepare 100ug/ml solution with 0.02M PBS buffer containing 0.5% BSA, then sterile filtered, filled and lyophilized to prepare the national standard candidate. The candidate was calibrated collaboratively using 2nd national standard as standards(150550-0004). The immune activity of candidate was 50ng/ampoule. Conclusion The candidate met the relevant requirements and can be served as the 3rd of national standard for quanti- tative immunoassay of T3. The establishment of the 3rd of national standard for quantitative immunoassay of T3 will provide a quantitative basis for quantitative immunoassay kits of T3.
