摘要 采用同步热分析(TGA-DTG-DSC)和热重-傅立叶红外(TGA-FTIR)联用技术研究齐多夫定(AZT)在氮气气氛和空气气氛中的热分解过程,测定AZT及其在热分解过程中不同阶段气体产物和固体残留物的红外光谱,运用量子化学GAMESS软件计算AZT分子的键级,探讨AZT的热分解机理.研究结果表明,AZT的热分解过程是一个三阶段过程,起始热分解步骤是联接胸腺嘧啶环与四氢呋喃环的C—N键的断裂,热分解中间残留物是胸腺嘧啶. The thermal decomposition process of zidovudine (AZT) was studied by means of thermo- gravimetry (TG), differential scanning calorimetry(DSC), and thermogravimetric analysis coupled with Fou- rier transform infrared spectroscopy(TGA-FTIR).The infrared spectra of AZT,its gaseous products and re- mainders from thermal decomposition at various temperatures were determined. The molecular bond orders were calculated by GAMESS method of quantum chemistry,and the mechanism of the thermal decomposition of AZT was discussed.The results indicated that the thermal decomposition of AZT was a three-stage process. Disconnection of the C-N bond between thymine ring and tetrahydrofuran ring occurred in the initial step of the decomposition, and the intermediate was confirmed as thymine.
