吡格列酮对糖尿病小鼠血脂、血糖的影响 被引量:1

摘要 目的观察吡格列酮对糖尿病小鼠血糖、血脂的影响,研究其可能作用机制。方法选取昆明种小白鼠30只随机分为三组:正常组、模型组、吡格列酮组,每组10只。采用一次性大剂量四氧嘧啶腹腔注射方法制作糖尿病小鼠模型,吡格列酮组给予吡格列酮灌胃,剂量为70 mg/kg,灌胃量为0.1 ml/10 g,1次/d,模型组和正常组灌胃相同体积的生理盐水。30 d后观察比较各组小鼠体重、进食量、饮水量、血糖、血脂的变化。结果模型组小鼠体重减轻,进食量、饮水量增加,血糖升高,吡格列酮组较模型组体重增加,进食量、饮水量减少,血糖降低。吡格列酮组TC(4.6±1.5)mmol/L,显著低于模型组(18.1±0.5)mmol/L(P<0.05);吡格列酮组TG(1.4±0.4)mmol/L,显著低于模型组(3.2±0.4)mmol/L(P<0.05)。吡格列酮组HDL-C(1.2±0.2)mmol/L,高于模型组(1.1±0.2)mmol/L(P>0.05)。结论吡格列酮对糖尿病小鼠有明显的治疗作用,能抑制其体重的下降,对进食量、饮水量等有明显的改善,能降低糖尿病小鼠的血糖,其作用机制可能与降低血中TG、TC,升高血中HDL-C等有关。这也可能是吡格列酮防治糖尿病并发症的机制之一。 Objective To observe the influence of pioglitazone on blood lipids,blood glucose in diabetic mice,and to study its possible mechanism.Methods Thirty KM mice were randomly divided into 3 groups:normal group,model group,pioglitazone group,with 10 mice in each group.One-time large doses by intraperitoneal injection of alloxan method was used to make diabetic model mice.Pioglitazone group was given pioglitazone,70 mg/kg,0.1 ml/10 g,1 time a day,while model group and normal group were given the same volume of physiological saline.After 30 days,the weight,food intake,water intake,blood glucose,blood lipids of each group were observed.Results In model group,mice's body weight reduced,food intake,water intake and blood glucose increased.In pioglitazone group,mice's body weight increased,food intake,water intake and blood glucose reduced,compared to those in model group.The TC was (4.6±1.5) mmol/L in pioglitazone group,and was significantly lower than (18.1±0.5) mmol/L in model group.The TG was (1.4+0.4) mmol/L in pioglitazone group,and was significantly lower than (3.2±0.4) mmol/L in model group.The HDL-C was (1.2±0.2) mmol/L in pioglitazone group mice,and was higher than (1.1±0.2) mmol/L in model group.Conclusion Pioglitazone has obvious therapeutic effect on diabetic mice,which can restrain the reduced weight,improve the food intake and water intake,and reduce the blood glucose of diabetic mice.The effect mechanism might be related to reducing the serum TG,TC,elevating serum HDL-C.
