摘要
目的研究microRNA-34a(miR-34a)对膀胱肿瘤细胞株5637细胞迁移、侵袭和增殖的影响。方法在5637细胞株中过表达miR-34a,运用小室迁移实验和小室侵袭实验检测细胞迁移力和侵袭力;运用血球计数板法和新型四唑氮盐(MTS)法检测细胞增殖力。结果过表达miR-34a后,5637细胞迁移力和侵袭力均下降(P<0.01),细胞增殖受抑(P<0.01)。结论过表达miR-34能抑制膀胱肿瘤细胞株5637的迁移、侵袭和增殖。
Objective To evaluate the effects of microRNA-34a （miRNA-34a） on cell migration,invasion and proliferation of bladder cancer cell line 5637.Methods miR-34a was overexpressed in bladder cancer cell line 5637.Transwell migration assay and transwell invasion assay were performed to predict cell migration ability and invasion ability,respectively.Cell proliferation was assessed by cell counting assay and MTS assay.Results The overexpression of MiR-34a significantly down-regulated cell migration and cell invasion of bladder cancer cell line 5637 （P〈0.01）,and significantly decreased cell proliferation （P〈0.01）.Conclusion The overexpression of microRNA-34a inhibits cell migration,invasion and proliferation in bladder cancer cell line 5637.
