摘要
目的观察川芎嗪对内毒素血症大鼠胸主动脉血管环对去甲肾上腺素收缩反应性的影响,并探讨血管平滑肌中iNOS、p65蛋白在其中的作用。方法 24只雄性SD大鼠分为四组:对照组、lps注射组、川芎嗪干预组、注射lps后川芎嗪治疗组。lps注射后6 h,应用体外血管条张力测定技术检测各组大鼠胸主动脉环对不同浓度去甲肾上腺素的收缩反应;应用免疫荧光法检测大鼠胸主动脉血管平滑肌中p65及iNOS蛋白的表达。结果与对照组相比,注射lps后大鼠胸主动脉血管环对去甲肾上腺素的收缩反应产生的张力显著降低;川芎嗪治疗后内毒素血症大鼠胸主动脉对去甲肾上腺素的反应性显著升高。内毒素血症大鼠胸主动脉血管平滑肌表达iNOS及p65蛋白较对照组显著升高,川芎嗪治疗后可抑制内毒素血症大鼠血管平滑肌中iNOS及p65蛋白的表达。结论川芎嗪可增强内毒素血症大鼠血管对去甲肾上腺素的收缩反应性,这与川芎嗪降低血管平滑肌中iNOS及p65蛋白表达有关。
Objective This study investigates the effects of Ligustrazine on thoracic vascular reactivity and the expression levels of p65 and iNOS in aortas in endotoxemia rats. Methods Twenty four male SD rats were divided into four groups:control group Lps injection group, ligustrazine treatment group and lps + ligustrazine treatment group. Six hours after lps challenge, vascular reactivity to norepinephrine of the thoracic rings were tested by in vitro vascular ring tension determine methods. The expression levels of iN- OS and p65 in thoracic smooth muscle were also tested by immunofluorescence. Results Compared with the control group, the rings from lps injection group had significantly lower response to NE. Lignstrazine treatment partially restored the response of tho- racic rings to norepinephrine. The expression levels of iNOS and p65 were significantly increased in lps injection group. Ligus- trazine treatment was able to decrease the levels of iNOS and p65 in endotoxemia rats. Conclusion Our results indicate that Ligustrazine was able to restore the vascular reactivity to norepinephrine in endotoxemia rats by decreasing the expression levels of iNOS and p65 protein in vascular smooth muscle.
